    tiger It is a now extinct marsupial and predator that lived in Australia New Guinea and Tasmania where the last specimen died in a zoo in 1936 The Tasmanian wolf was striped had a long tail and was rather dog like The animals whose DNA has now been studied lived in zoos in Washington D C and London and were preserved at the Smithsonian Institution and the Swedish National Museum of Natural History in Stockholm respectively It was fascinating to discover that the two animals were so similar The genetic variation between them was very small says Anders Götherström a researcher at Uppsala University who together with his colleague Love Dalén initiated the project and was responsible for the DNA extraction and some of the phylogeny work among other things Little genetic variation is characteristic of animals that are threatened with extinction today and now we can see the same thing in the Tasmanian wolf more than 70 years after it died out According to these scientists too little genetic variation can lead to among other things a limited ability to withstand bacterial infections or other stress from the environment The DNA that was analyzed is mitochondria DNA extracted from the hair of the museum specimens Besides the Tasmanian wolves own DNA the researchers found DNA from other organisms including microorganisms that proved to be very different depending on what conservation technique had been used One of the animals was preserved in ethanol Stockholm and the other was kept dry as a hide in room temperature Washington These findings tell us a great deal about bacterial attacks after an animal has been prepared for museum purposes and in the long run can lead to better techniques for enhanced preservation There has been a great deal of interest in examining the

    ancient mitochondrial DNA Furthermore several properties of shafts suggest that they constitute an attractive DNA source for SBS sequencing by synthesis First their relative abundance when present renders them preferable to bones because the destructive nature of bone sampling can lead to the loss of important morphological information Second turnover of keratinocytes in the hair bulb is exceedingly high second only to that of cells in the gut epithelium Therefore baseline mitochondrial levels in these cells and thus the precortical cells that develop into the bulk of the shaft may be higher than those in other tissues commonly used for ancient DNA analyses Third even when degraded shafts are resistant to contamination from exogenous DNA such as bacteria blood and skin cells Our earlier work showed that hair shafts are a good source of mitochondrial and nuclear DNA from woolly mammoths However for the following reasons it remained to be tested whether hair shafts work well for sequencing other species Mammoth remains are preserved in the permafrost and DNA damage is known to increase rapidly with a specimen s post mortem temperature Mammoth hairs are larger and thus presumably more protective than hairs from some other species Mammoth hair can

    not know the full genome sequence of any immediate relative In contrast when sequencing the woolly mammoth we could compare our data with the available sequences from the African savanna elephant But for the thylacine the closest sequenced genome is from a South American marsupial called the short tailed opossum Latin name Monodelphis domestica The two lineages have been separated at least since the land bridge between South America and Australia was broken which happened perhaps 60 million years ago Mammoths and elephants have been separated only about 6 million years The 60 million years of separation together with the small sizes of many of the sequence fragments that we generated made it difficult to be certain that a particular DNA fragment was actually from the thylacine as opposed to being say human contamination To improve the odds we used the fact that for nuclear genome sequences that encode a protein the similarity between the thylacine and Monodelphis sequences will almost always be much higher than for an arbitrary region of the genome In summary our strategy was to see how much of the thylacine sequence aligned to Monodelphis protein coding intervals and then extrapolate to the full genome See
